FOXO6 (Forkhead Box O6) encodes a brain-enriched transcription factor of the FOXO family, which regulates genes involved in neuronal development, synaptic plasticity, memory formation, and oxidative stress response.[1] Unlike other FOXO family members (FOXO1, FOXO3, FOXO4) that are widely expressed, FOXO6 shows preferential expression in hippocampus and cerebral cortex, where it plays critical roles in learning and memory.[2] Dysregulation of FOXO6 signaling is implicated in cognitive decline and Alzheimer's disease pathogenesis.
The FOXO6 gene spans approximately 12 kb on chromosome 1p33 and contains 3 exons.[3] The encoded 524-amino acid protein contains:
Expression in the nervous system: FOXO6 is highly enriched in the hippocampal formation (CA1, CA3, dentate gyrus), with significant expression in cerebral cortex, amygdala, and olfactory bulb.[4] Expression increases during postnatal development and peaks in adulthood.
